Bei der Einstufung ist zu berücksichtigen, ob der Stoff auf bestimmten Expositionswegen resorbiert wird oder nicht, ob es bei den geprüften Expositionswegen nur zur lokalen Tumorbildung am Applikationsort kommt und ob bei geeigneten Prüfungen anderer wichtiger Expositionswege die karzinogene Wirkung ausbleibt. The classification shall take into consideration whether or not the substance is absorbed by a given route(s); or whether there are only local tumours at the site of administration for the tested route(s), and adequate testing by other major route(s) show lack of carcinogenicity.

Substances which have induced benign and malignant tumours in well performed experimental studies on animals are considered also to be presumed or suspected human carcinogens unless there is strong evidence that the mechanism of tumour formation is not relevant for humans.

Der Gemeinsame FAO/WHO-Sachverständigenausschuss für Lebensmittelzusatzstoffe (JECFA) kam auf seiner 65. Tagung vom 7. bis 16. Juni 2005 zu dem Schluss, dass Acetamid (FL 16.047) bei Mäusen und auch Ratten eine eindeutig karzinogene Wirkung aufweist, und dass, obwohl der Mechanismus der Tumorbildung nicht bekannt ist, ein genotoxischer Mechanismus nicht ausgeschlossen werden kann. The Joint FAO/WHO Expert Committee on Food Additives (JECFA) concluded during its 65th meeting of 7 to 16 June 2005 that the acetamide (FL 16.047) is clearly carcinogenic in both mice and rats, and although the mechanism of tumour formation is unknown, the possibility of a genotoxic mechanism can not be discounted.
